Bermain piano sejak usia enam tahun, dilanjutkan dengan mempelajari piano klasik selama satu setengah tahun, disusul dengan teori klasik dan komposisi klasik, di mana setelah lulus ia menerjunkan diri untuk mengajar, semuanya ini ia tekuni di Manhattan School of Music. Tahun 1977 ia bergabung dengan pemain biola Michal Urbaniak untuk membantu pada kibord dan sinthesizer dalam turnya ke Eropa dan Skandinavia. Sekembali ke AS, ia bermain serabutan, di antaranya bersama Angela Boffill dan Don Alias. Mulai tahun 1979, untuk dua tahun lamanya ia bermain untuk trompetis Jepang, Terumasa Hino dalam tur Jepangnya.

Di Jepang inilah ia bertemu dengan trompetis muda pendobrak dunia musik jazz, Wynton Marsalis. Lepas Terumasa, mulai tahun 1982 hingga 1984 ia pun bermain untuk Wynton, baik dalam rekaman maupun untuk tur dunianya. Setelah sebelumnya ia mendampingi Jim Hall, Eddie Gomez dan Grady Tate untuk konsert di Jepang, tahun 1985, bersama-sama bassist Darryl Jones, drummer Omar Hakim dan saxophonist Branford Marsalis, Kenny bergabung dengan raksasa pop-rock asal Inggris, Sting. Ia mengaku banyak dipengaruhi oleh Herbie Hancock, Kenny Baron dan Larry Willis (pianis kelompok Blood Sweat & Tears).

Kenny Kirkland meninggal secara tragis akibat kegagalan jantung bulan September, 1998 di apartemennya di Bronx, New York.
